How It Works
Tirzepatide is a novel dual incretin agonist that activates both GIP and GLP-1 receptors, the two primary gut hormones responsible for post-meal insulin secretion. The GIP component enhances fat metabolism and may improve the tolerability of GLP-1-mediated appetite suppression. In head-to-head trials, tirzepatide produced up to 22.5% mean body weight reduction, exceeding semaglutide, and delivered superior HbA1c lowering in patients with type 2 diabetes.

Safety & Considerations
Side effect profile similar to GLP-1 agonists: nausea, diarrhea, vomiting, and constipation are most common and generally transient with dose escalation. Same boxed warning as semaglutide for thyroid C-cell tumors in rodents. Contraindicated in patients with MEN2 or personal/family history of medullary thyroid carcinoma. Monitor for pancreatitis, gallbladder events, and hypoglycemia when combined with insulin or sulfonylureas.
